* 12 Week Capros Trial Overview
Reducing Systemic Inflammation – Reducing Oxidative Stress – Increasing Nitric Oxide
- During this 12-week trial, Capros showed significant improvement in endothelial function, oxidative stress, immune response, and lipid profile.
- Out of the 65 screened subjects, 59 eligible subjects were randomized and completed the study.
- The study consisted of two different dosages of Phyllanthus emblica extract (Capros) – 250 mg and 500 mg twice daily. Both dosages showed significant improvement in endothelial function (cardiovascular health), oxidative stress biomarkers-Nitric oxide (+ 41.89%, + 50.7%), Glutathione (+ 24.31%, + 53.22%), Malondialdehyde (- 21.02%, – 31.44%), and systemic inflammation biomarker, hsCRP (- 39.68%, – 53.77%) at 12 weeks with 250 mg and 500 mg twice daily dosage respectively, compared to baseline and placebo.
- Results also showed a significant mean percent change at 12 weeks with total cholesterol (- 7.71%, – 11.11%), HDL-C (+ 7.33% + 22.16%), LDL-C (- 11.39%, – 21.8%) and Triglycerides (- 9.81%, – 19.22%) respectively with 250 mg and 500 mg twice daily. The 500 mg twice daily dose was significantly more efficacious than the 250 mg twice daily dose and placebo.
Healthy Immune Response
Capros Amla is standardized to potent polyphenols, including hydrolysable tannins, emblicanin A, emblicanin B, punigluconin and pedunculagin. Polyphenols are powerful antioxidants found in a variety of plant extracts that have been shown to support cognitive health, digestion, heart health and a healthy immune response. These polyphenols have been shown to support the immune system by modulating ROS and lipid peroxidation. Tannins, found in Capros, have been shown to impact ROS, MDA, antioxidant GSH and cytokine production. Polyphenol bioavailability, including tannins, is typically very low due to limited absorption, extensive metabolism and rapid excretion. Capros’ unique aqueous extract preserves the polyphenols and at 500 mg (250 mg twice a day) ensures cardio benefits indirectly supporting a healthy immune response. In addition to heart health, Capros further supports the body’s natural balance by protecting the body against oxidative stress and supports a healthy immune response without being a pro-oxidant.
Cardiovascular Health
Acute and chronic stress is a risk factor for the development and progression of coronary artery disease. Increased arterial stiffness is an independent marker for cardiovascular disease. Cold pressor test (CPT) is known to be associated with substantial activation of the autonomic nervous system.
Objective
The aim of this study was to evaluate the effect of Phyllanthus emblica extract (Capros) on cold pressor stress test induced changes on cardiovascular parameters and aortic wave reflections in healthy human subjects.
Materials and Methods
- This was a double-blind, placebo-controlled, crossover study. Participants were randomized to receive either two capsules of P. emblica extract 250 mg (containing aqueous extract of P. emblica, highly standardized by high-performance liquid chromatography to contain low molecular weight hydrolysable tannins emblicanin-A, emblicanin-B, pedunculagin and punigluconin) or two capsules of placebo twice daily for 14 days.
- Pharmacodynamic parameters such as heart rate, augmentation pressure, augmentation index (AIx), subendocardial viability ratio (SEVR), radial and aortic blood pressure (BP) were recorded before and after CPT at baseline and end of treatment.
- After a washout period of 14 days, subjects crossed over to the other treatment and the same test procedure was repeated again. Safety assessments were done at baseline and at the end of treatment.
Results
A total of 12 volunteers completed the study. Compared with baseline and placebo, P. emblica extract produced a significant decrease of mean percent change in the indices of arterial stiffness (AIx, radial and aortic BP) and increase in SEVR, an index of myocardial perfusion with CPT. Both treatments were well-tolerated, and no serious adverse events were reported.
Mental Stress Management
Introduction
Experimental studies have found that acute stress leads to pathophysiological changes in cardiac risk profile and thereby more directly explain a link to cardiovascular disease. The present study was undertaken to evaluate the effect of Emblica officinalis (Capros) versus placebo on mental stress induced changes in cardiovascular hemodynamic parameters and arterial wave reflection properties in healthy human subjects.
Materials & Methods
In this randomized, double-blind, placebo-controlled, crossover study, participants received either two capsules of standardized Emblica officinalis fruit extract 250 mg or placebo daily for 2 weeks, as per prior randomization schedule. Pharmacodynamic parameters were recorded before and after a standardized mental stress test, at baseline and at the end of treatment. After a 2-week washout, participants crossed over to receive the other treatment.
Results
A total of 12 volunteers completed the study. Compared to baseline, Emblica officinalis fruit extract produced a statistically significant decrease in Aortic augmentation pressure (from 6.6±2.15 to 4.6±2.02); (p< 0.01) and in Augmentation Index AIx (from 120.1±12.80 to 116.2±12.63); (p< 0.05). There was a reduction in Radial and Aortic systolic (SBP) and diastolic (DBP) pressures and an increase in Sub Endocardial Viability Ratio (SEVR), which was however not statistically significant.
Conclusion
Acute mental stress results in temporary increase in arterial stiffness and wave stiffness. Standardized Emblica officinalis fruit extract decreased the mental stress induced changes on aortic wave reflections in normal healthy subjects. These results suggest the beneficial effect of Emblica officinalis in mitigating the effects of stress, thereby reducing the risk of acute cardiovascular morbidity.
